John was born in Damascus, Syria, to a wealthy Christian family. His father, Mansur ibn Sarjun, was the chief financial officer of the Umayyad Caliph Abd al-Malik ibn Marwan. John received a comprehensive education in Greek philosophy, rhetoric, and Christian theology. He also studied Arabic, possibly due to the influence of his Arab environment. Despite his privileged upbringing, John chose to pursue a life of asceticism and scholarship, becoming a monk at the Monastery of Mar Saba near Jerusalem.

John's theological writings were heavily influenced by the Greek Fathers of the Church, particularly Basil the Great, Gregory of Nazianzus, and Maximus the Confessor. He synthesized their teachings with the philosophical insights of Aristotle and Neoplatonism, creating a unique and comprehensive system of Christian thought. John's most famous work, the "Exposition of the Orthodox Faith," is a systematic presentation of Christian doctrine that became a standard textbook in Eastern Christianity.

In addition to his theological writings, John also composed numerous hymns that are still sung in Eastern Orthodox and Catholic liturgies today. John's hymn "O Gladsome Light" is particularly well-known and is sung at the beginning of every Orthodox Vespers service.
